There are no known drugs or drug combinations that promote substantial central nervous system axonal regeneration after injury. We used systems pharmacology approaches to model pathways underlying growth and identify a four-drug combination regulates multiple subcellular processes in the cell body axons using optic nerve crush rats. intravitreally injected agonists HU-210 (cannabinoid receptor-1) IL-6 (interleukin 6 receptor) stimulate retinal ganglion cells for growth. applied, gel foam at site of injury, Taxol stabilize growing microtubules, activated protein C clear debris field since computational models predicted this regulating two cone produces synergistic Physiologically, treatment restored preserved pattern electroretinograms some animals had detectable visual evoked potentials brain behavioral optokinetic responses.
